Public interest in probiotics is on the upswing, if the glut of advertisements for probiotic yogurts?those with added doses of beneficial bacteria?is any evidence. Scientists are bringing this traditional therapy into the 21st century by genetically engineering the microbes to enhance their effect on the immune system. They hope the new bugs will ultimately help treat inflammatory bowel diseases such as Crohn's disease and ulcerative colitis, as well as other disorders that result from an overactive immune system.
In research published today in the Proceedings of the National Academy of Sciences, scientists deleted a gene from the bacterium Lactobacillus acidophilus, which is commonly found in yogurt. Mansour Mohamadzadeh, associate professor of medicine at Northwestern University, and collaborators had previously shown that the enzyme this gene manufactures increases inflammation, a defining characteristic of Crohn's disease and ulcerative colitis. But the unaltered form of the bacterium also triggered production of a beneficial immune molecule, IL-10m, which helps to regulate the immune system. The goal of the engineering the microbes was to deliver the beneficial effects without the harmful ones.
When fed to mice with colitis and inflammation of the colon, the engineered bacteria prevented the weight loss and bloody diarrhea that typically accompanies this condition. In addition, the treated mice had 90 percent less inflammation in their colon tissue than did their untreated counterparts.
While probiotic foods and supplements are a huge industry, it's unclear whether they actually help treat most gastrointestinal diseases. The research published today is part of a trend in microbiology to explore in rigorous detail the effects of probiotics and the mechanisms that underlie them.
"The concept [of probiotics] is wonderful, but the evidence of their [effectiveness] is fairly limited," says Balfour Sartor, co-director of the Center for Gastrointestinal Biology and Disease at the University of North Carolina, who was not involved in the new study. Because probiotics are considered a food and not a drug, they are not regulated by the U.S. Food and Drug Administration, and therefore don't require the large clinical tests that drugs do.
Inflammatory bowel disease is one of the prime areas of interest for probiotic treatment, but "there really has been little direct evidence that probiotics are effective in treatment or prevention of Crohn's disease," says Sartor. Some research suggests that two different probiotic formulations can help prevent recurrence of ulcerative colitis, he says. "But that's just two out of thousands of formulations."
Scientists still don't know exactly how probiotic bacteria influence the gastrointestinal system, but previous research suggests several possible mechanisms. Beneficial bacteria might temporarily alter the ratio of good to bad bacteria that inhabit the intestine, or they might specifically block activity of bad bacteria. And probiotics seem to influence the immune system, "stimulating protective immune cells or blocking detrimental activities of immune cells," says Sartor.

Mr. Huntsman, a former Republican governor of Utah, was nominated nearly two years ago by President Obama to serve as the top American envoy to China. He has not decided whether to move forward with a candidacy, associates said, but he has had several conversations with a circle of political advisers who are waiting in the wings if he decides to run.
While Mr. Huntsman has declined to discuss his political ambitions, particularly those that could include challenging the president for whom he has served, a trail of evidence has been building that signals his interest in a White House bid. He and his family have told friends that they are eager to leave Beijing and come to Washington, where he bought a home for $3.6 million last summer.
The first Republican contests of the 2012 nominating season are one year away and at least a half-dozen candidates are seriously surveying the landscape. It remains an open question how Mr. Huntsman would appeal to Republican primary voters, particularly because of a moderate record he assembled in his four years as governor of Utah, where he became a leading moderate voice of the party on issues like immigration, gay rights and the environment.
And, of course, the fact that Mr. Huntsman, 50, worked in the Obama administration could also be a complicating factor. Asked about his ambassador?s becoming a potential political rival, Mr. Obama resorted to humor at a January news conference with President Hu Jintao of China.
?I couldn?t be happier with the ambassador?s service, and I?m sure he will be very successful in whatever endeavors he chooses in the future,? Mr. Obama said, laughing as he discussed the political future of Mr. Huntsman. ?And I?m sure that him having worked so well with me will be a great asset in a Republican primary.?
Mr. Huntsman, who supported Senator John McCain?s presidential bid in 2008, began quietly reaching out to political advisers late last year to explore a 2012 candidacy. The political team for Mr. Huntsman, which was first reported last week by The Washington Post, includes John Weaver, a former adviser to Mr. McCain, and Fred Davis, who produces television ads for Republican candidates. Mr. Huntsman has told associates that he will make a decision by June.
The subject of Mr. Huntsman?s political ambitions was raised Monday at the White House news briefing, where Robert Gibbs, the president?s press secretary, confirmed that Mr. Huntsman had informed administration officials of his plans to step down and that a search was under way for a new ambassador to China.
?The president and, I think, the American people expect that somebody that holds the post of ambassador from the United States to China will dedicate their full energy and time to that position,? Mr. Gibbs said. ?And we believe that Ambassador Huntsman believes that as well.?
Mr. Huntsman?s decision to leave after nearly two years of service is neither unexpected nor without precedent. Ambassadors often hold their positions for only part of the president?s term. When he accepted the position, standing in the Diplomatic Reception Room of the White House on May 16, 2009, Mr. Huntsman expressed his surprise.
?Nor did I expect, as national co-chair of Senator McCain?s presidential campaign, to be called into action by the person who beat us,? Mr. Huntsman said. ?But I grew up understanding that the most basic responsibility one has is service to country.?

Anyone noticed this? I thought I might have accidentally retweeted myself earlier but after it happened four times that just isn?t a possibility.
Initially I believed it to be a Tweetdeck bug (after its update yesterday) but after reaching out to the team, they reported that it?s Twitter itself with the same issue across all twitter clients.
Visiting a twitter profile page won?t show any of the retweets but twitter searches do reveal them.
What?s frustrating about this you ask? I keep getting retweet notifications for things I said(!)
We?ve contacted twitter and will update if/when we hear back.
Update: We?ve discovered that the retweets only appear for searches in your twitter client, not for mentions. So if you happen to have a search set up for you own name and want a temporary solution, just switch to mentions for now.

As we reported yesterday, a major initiative to boost the American economy by promoting entrepreneurship has been announced with a wide range of partners including Facebook, IBM and Intel.
It won?t just be US citizens that will benefit from Startup America, though. Three European accelerator schemes have signed up to join a network that will give them better access to the American startup scene.
The UK?s Springboard, Propeller from Ireland?s Ryan Academy for Entrepreneurship, and Startup Bootcamp, which has bases in Spain and Denmark, have all joined the TechStars Network. Launched yesterday by US accelerator scheme TechStars, the network largely consists of American programmes, although the three European schemes are joined by Singapore?s JFDI Asia in representing the rest of the world.
The TechStars Network?s target is that over the next four years, 5000 experienced business leaders and investors will mentor and support 6000 promising young entrepreneurs, with the aim of increasing the success rate and sustainability of each program and creating 25,000 new jobs by 2015.
Springboard?s Jon Bradford said in the announcement of its involvement, ?Being part of the TechStars Network gives us greater access to a wider network of mentors and the provision of a wide alumni for the start-ups and programme organisations involved. It will also accelerate best practice for mentorship-drive startup programmes and we hope this opportunity will inspire European startups to take part.?
TechStars founder David Cohen will be speaking about the Network?s launch at Springboard?s ?Meet the Mentor? event, which takes place this Thursday, 3 February at TechHub in London.

Swedish startup Twingly?s Channels product (which impressed us at launch) is essentially a ?FriendFeed for the enterprise market? and now the company is stepping onto Salesforce Chatter?s turf with the launch of a Twitter-like corporate microblogging service.
Although he describes the move as a ?pivot?, Twingly CEO Martin Källström says that this change in focus isn?t down to a failing business model. In fact, he says that the startup is now breaking even, after having made a major sales drive during 2010. Instead, he says that the existing Channels product simply wasn?t effective enough for group communication.
?After looking at how people use Twingly, we saw that the conversation really needed to be in a separate track from the aggregated links, so that it isn?t ?washed away? by content from feeds and search. We realized that focusing on the conversation takes us further in the direction we want to go in the long run. We can provide more value by allowing people to meet other people than by reading a stream of news on their own.?
While news aggregation and blog search are still part of Twingly?s offering, the default view once logged in is a basic microblogging service. Källström says that he?s not directly targeting established players in the space, instead looking at how Twingly can offer something different to Yammer and Salesforce Chatter. ?We are exploring what group communication tools we can develop that can be used alongside Yammer, like the open discussions now available on Twingly.com.?
While customers of other corporate microblogging providers are unlikely to switch to Twingly?s service purely for that part of its service, it?s easy to see how companies involved in PR and other activities that require a good understanding of what people are saying in the outside world could find a use for this ?private Twitter meets FriendFeed? experience.
